The Human serum albumin (HSA) used is Sigma A-3782 (essentially fatty acid free. Prepared from crystallized and lyophilized essentially globulin-free albumin).
The incorporation of biotin to the albumin is done by reating Sulfo-NHS-LC-Biotin (Pierce, 21335) with the albumin at neutral pH. The primary amines of the protein react with the NHS-esters on biotin to form stable amide linkages to the protein.
The incorporation of biotin to the albumin is measured by time of flight mass spectrometry and shows 28 mole of biotin per mole protein.
